Do the S-Video cables really make that much of a difference?

When compared to composite...

When compared to composite... yes. Composite connections carry the luma and both chroma signals mixed together. The display has to seperate them with a comb filter, which causes numerous types of picture anomalies. 

S-Video connections carry luma and chroma signals seperately, which drastically improves image quality. When dealing with analog sources, component video offers the best performance, as it keeps all three signals seperate on their own cables. The difference between component and S isn't as drastic with 480i analog material, but every little bit helps!
